Simoncelli memulai karir di tahun 2002 di kelas 125 World Championship. Sepanjang empat tahun berlaga di kelas ini, pembalap yang bergabung di tim Aprilia ini sukses menang sebanyak tiga kali dan tujuh kali naik podium dari 50 kali tampil.
Pada tahun 2006, Pembalap yang akrab disapa Super Sic ini naik kelas ke 250 World Championship. Bersama tim Gilera, sepanjang keikutsertaannya di kelas ini, Simoncelli berhasil memenangi 12 laga dan 22 podium dari 64 serie yang diikutinya.

Simoncelli berhasil menjuarai 250 World Championship pada musim 2008 dengan memetik 280 poin. Di musim itu, ia mampu memenangi 6 seri balapan. Namun sayangnya, di musim selanjutnya, ia tidak dapat mengulangi suksesnya. Simoncelli harus puas berada di urutan ketiga klasemen akhir 2009.

Meski demikian, kepiawiannya mengendarai 'kuda besi,' membuat Honda tertarik kepadanya. Simoncelli pun naik kasta pada 2010 dengan bermain di kelas MotoGP World Championship bersama Honda. Di musim pertamanya, Simoncelli hanya mampu berada di posisi ke delapan dengan perolehan 125 poin.

Sementara itu, di musim ini sendiri, posisi Simoncelli masih terpaku di posisi keenam dengan perolehan dua kali menaiki podium dari 16 laga musim ini.
